Abstract
Objective:In order to obtain natural nucleosides with higher purity,the separation and enrichment process of nucleosides in Pinctada martensii was studied. Methods:The nucleosides from the liquid decocted with water and deposited with alcohol of Pinctada martensii were used as indicators to determine the type of macroporous resin by HPLC-UV,the loading process and the elution process.The nucleoside at the purified site was determined by UV. According to the results obtained by HPLC-UV,the selected macroporous resin model was SP207,the loading concentration was 2.19 mg/mL,the loading pH was 4.6,the loading flow rate was 2 BV/h,and the optimal loading volume of nucleoside was 25 mL. The elution time was 2 BV,the elution solvent was 15% ethanol,the elution flow rate was 3 BV/h,the elution volume was 3 BV,and the aspect ratio was 1:7. Conclusion:After SP207 purification,the purity of total nucleosides was 58.19%,which indicated that this method was suitable for the separation and enrichment of nucleosides in Pinctada martensii.
