Determination of Total Sugar and Reducing Sugar in Degreasing Cream from Periplaneta americana

  • Objective:The content of polysaccharides in the degreasing cream of Periplaneta americana was determined by different methods in order to select an accurate and convenient method for determination of total sugar content in Periplaneta americana extract and establish a method for determining reducing sugar content. Methods:Phenol-sulfuric acid method and anthrone colorimetry were used to determine the total sugar content. DNS method was used to determine the reducing sugar content. Results:The glucose was in a good linear relationship in the range of 0.004~0.100 mg/mL (R2=0.9996), and the recovery rate was 98.7% (RSD=2.0%) by phenol-sulfuric acid method. The glucose was in a good linear relationship in the range of 0.02~0.20 mg/mL (R2=0.9997), and the recovery rate was 97.7% (RSD=2.6%) by anthrone colormetry. The glucosewas in a good linear relationship in the range of 0.004~0.040 mg/mL (R2=0.9996), and the recovery rate was 96.8% (RSD=1.6%) by DNS method. Conclusion:The phenol-sulfuric acid method could be used to determine the total sugar content. The DNS method could be used to determine the reducing sugar content.
