Abstract
Zizyphi Spinosae Semen polysaccharide decoloration process was optimized by the method of response surface optimization. Firstly,the resin type with a well decolorization process was selected by static adsorption method. Then the sample concentration(mg/mL),sample volume(mL)and elution velocity(BV/h)were used as the main factors in the process optimization by single factor experiment. Based on response value(OD)of polysaccharide decolorization rate and retention rate,response surface method(RSM)was used to analyze optimization the decolorization process by Box-Behnken design(BBD). The results showed that the optimized decolorization parameters with D101 macroporous adsorption resin were 2 mg/mL for the sample concentration,3.5 mL for the volume,and 4 BV/h for the elution flow rate. Under the condition,the value of polysaccharide decolorization rate and retention rate were 61.32% and 87.05%,respectively,and the OD value was 0.56(the predicted value of 0.55),which indicated the optimized decoloration method was feasible.
