Preparation of Salmonella and Vibrio parahaemolyticus Co-enriched Culture Medium

Abstract
To prepare a culture medium which could simultaneously increase Salmonella and Vibrio parahaemolyticus, the components of specific culture medium was based on GB 4789.4-2016 and GB 4789.7-2013, different nutritional requirements of the target bacteria were taken as references to select suitable accelerators and inhibitors. The culture medium components were determined and the enriching effects were verified in this study. Results showed that, the components of Salmonella and Vibrio parahaemolyticus co-enriched culture medium as follows:Peptone 10.0 g, potassium dihydrogen phosphate 1.5 g, sodium chloride 7.5 g, sodium thiosulfate 5.0 g, cattle bile salt 0.1 g, sodium citrate 2.5 g, mannitol 2.5 g, glucose 2.5 g, distilled water 1000 mL. The initial inoculum concentration of the target bacteria both were 102 CFU/mL, and the concentration could reach to 107~108 CFU/mL after culturing at 37℃ for 16 h. The results showed that the co-enriched culture medium could be used for the enrichment of Salmonella and Vibrio parahaemolyticus. The culture medium could be used for the enrichment and cultivation of Salmonella and Vibrio parahaemolyticus, which was easy to be prepared, cost-effective, and had a good market prospect.
