Optimization for extraction of polysaccharides from Apostichopus japonicus spawn using double enzymetic by response surface methodology and antioxidant activity analysis

Abstract
On the basis of single factor experiments,the double enzymetic extraction process was optimized to extract polysaccharides from Apostichopus japonicus spawn with response surface methodology,and the preliminary antioxidant activity of polysaccharides was investigated. The results showed that the best optimum condition for the extraction of polysaccharides were as follows: the ratio of protamex and papain was 1∶ 2,enzyme dosage 2.78%,enzymolysis temperature 60 ℃,enzymolysis time 6h. Under such condition,the model- predicted and experimental values of polysaccharides yield were 8.84‰ and 8.76‰,respectively.Polysaccharides had ability to scavenge free radical in a dose- dependent manner,the IC_(50) values of DPPH free radical scavenging abilities were 1.71 mg / m L.The A.japonicus spawn polysaccharides is a good source of natural antioxidants.
