Determination of 6-BA and its residual dynamics in bean sprouts by SPE-UPLC

Abstract
This study was aimed to set up a novel method to detect the residual content of 6-Benzylaminopurine(6-BA) of three kind of bean sprouts with the solid-phase extraction-ultra performance liquid chromatography and try to indicate the dynamic variation of content of 6-BA in bean sprout with different storage conditions and interval times. DIONEX Acclaim C16( 4.6 mm ×150 mm, 3 μm) was used and the mobile phases were methanol and ammonium acetate,respectively,with a ratio of 45% methanol. Flow velocity was 1 m L·min-1and the detection wavelength was 265 nm. The results showed that spiked recoveries were 87.4%~90.0%. The relative standard deviation was 0.46%~0.60%. Detection limit and linear range of 6-Benzylaminopurine was about 0.01 mg·kg-1and 0.05~50 mg·kg-1respectively. The novel method was quite easy to use and could be used to detect accurately the content of 6-BA. The content of 6-BA showed the drop trend with the rise of storage time and the minimum half-life period was 1.642 and 3.108 d respectively under the conditions of frozen storage and cold storage.
