Abstract
Yak blood as the research object, the prothrombin was prepared by adsorption method with barium citrate. The study optimized preparation conditions of Ca2 +activating the prothrombin in yak blood by single factor experiment (the activating temperature, final concentration of Ca2+and activating time) and Box-Behnken experiment. The results showed that the optimal thrombin specific activity was 63.3972U/mg at the following conditions:activating temperature 27℃, final concentration of Ca2+0.01mol/L, activating time 1.95h.
