Study on optimization of SOD extraction conditions from wheat seedlings

Abstract
The phosphate buffer was used to extract SOD, the antioxidant component of wheat seedlings. Four key extraction process parameters factors selected for single-factor tests were phosphate buffer pH value, extraction time, heat treatment temperature, and the heat treatment time. Four factors and three levels of response surface analysis (RSA) was used to determine the optimum levels of four factors above. Then activity of the SOD was measured by adjacent benzene three phenolic autoxidation method. RSA results indicated that the optimal levels of these parameters were as follows:phosphate buffer pH value 8.0, extraction time 1h, the heat treatment temperature 70℃ and the heat treatment time 15min. Under these conditions, the activity of the SOD was up to 587.60U/mL. This study provided a new direction for the comprehensive utilization of wheat seedlings.
