Study on separation and identification scotch pine bark polyphenols with HPLC method

  • The HPLC method was employed to separate and determine polyphenols in park of pinus sylvestris var. mongolica. To optimize the separation conditions, detecting wavelength, mobile phase, mobile rate, column temperature and injecting volume were investigated. The optimal chromatographic separating condition was established: the detecting wavelength was 280nm, mobile phase was methanol-water (0.05% TFA) , mobile rate was 1.0mL/min, column temperature was 30℃ and injecting volume was 5μL. Through comparison with standard sample chart, p-Coumaric acid, catechin, caffeic acid, ruitin, chlorogenic acid, gallic acid, cinnamic acid was detected in park of pinus sylvestris var.mongolica.The high content of p-Coumaric acid and catechin in park of pinus sylvestris var.mongolica were analyzed.
