Determination of hydroxymethylfurfural in pomegranate juice concentrates by solid phase extraction-ultra performance liquid chromatography

  • An ultra performance liquid chromatography (UPLC) method had been developed for the determination of hydroxymethylfurfural (HMF) in pomegranate juice concentrate.Samples were diluted with water, and then cleaned up with a Bond Elut ENV solid phase extraction cartridges.UPLC was performed on a UPLC HSS T3 (100mm×2.1mm, 1.8μm) using acetonitrile-water (8+92, V/V) as mobile phase.The detection wavelength was 285nm.Quantitation was carried out using an external standard method.The detection limit of 5-HMF was 0.02mg/L.The results showed good linear relationship at the range of 0.1~10.0mg/L.Recoveries spiked at 10、 20、50mg/kg ranged from 80.8%to 110.8%with relative standard deviations (RSD) less than 8.1%.The method was simple, fast and accurate and could be applied to the analysis of 5-HMF content in pomegranate juice concentrate.
