Optimization of extraction conditions of polysaccharide from loquat core by response surface methodology

Abstract
Response surface methodology (RSM) was applied to optimize the extraction conditions of loquat core polysaccharides.Based on single factor experiment, the amount of cellulase, pH, enzymolysis temperature and reaction time were chosen as influencing factors, the loquat core polysaccharides yield was selected as response value.The mathematical model was established by Box-Behnken central composite design.The results showed that the optimal extraction conditions were as follows:material-water ratio 1:15, ultrasonic temperature 60℃, ultrasonic time 30min, the amount of cellulase 1.8%, enzymolysis temperature 49℃, enzymolysis time 1.8h and pH4.7.In this optimal process condition, the yield of loquat core polysaccharides was 9.96%.
