Abstract
Objective:To study the optimal separation and purification technological parameters of peach blossom polyphenols with macroporous resins.Methods:6 types of macroporous resins were compared through static experiments.D-101 resin was screened out for higher adsorption and desorption performances, and then its dynamic adsorption-desorption properties were investigated to get the optimum parameters.Results:D-101 resin was suitable for purifying peach blossom polyphenols.The optimum technology parameters were as follows:4.5BV sample added on the column, and the sample concentration was about 0.744mg/mL at the flow rate of 1BV/h;then washed by deionized water to the outflow nearly colorless, the desorption was eluted with 80% alcohol at flow rate of 1BV/h, the eluant volume was about 3BV.The peach blossom polyphenols purity increased from 6.17% to 29.30%±3.04% and the total polyphenols yield reacheed 79.01%±3.39%, under the optimum conditions.Conclusion:The method was simple and feasible with good effect of separation, which could meet the industrial requirements.
