Analysis of compositions of anthocyanins in wild mulberry

Abstract
To establish a method to determine anthocyanin content and composition in wild mulberry, solid phase extraction (SPE) and high performance liquid chromatography equipped with a DAD detector and electro-spray ionization mass spectrometry (HPLC/DAD/ESI/MS) were used.The analysis was carried on Zorbax SB-C18 column (250mm×4.6mm, 5μm) .The mobile phase was aqueous 5% formic acid solution and methanol at the flow-rate of 1.0mL/min, the detection wavelength was 520nm.The content of gross anthocyanins was 154.27mg/100g by HPLC expressed as milligrams of cyanidin 3-glucoside equivalent per 100g of fresh weight.Three anthocyanins in wild mulberry were identified by UV-Vis and ESI+-MS spectra which were cyanidin 3-glucoside, cyanidin 3-rutinoside and pelargonidin 3-glucoside, the relative content of which were 67.52%, 31.29%, 1.06% respectively.
