摘要: 利用响应面分析法优化八宝景天叶中总黄酮的超声提取工艺。以总黄酮提取率为评价指标,在考察单因素实验的基础上,利用Box-Behnken响应面分析法确定最佳提取工艺条件。八宝景天叶中总黄酮的最佳提取工艺条件为:乙醇浓度72%,液料比21:1mL/g,超声温度为64℃,超声时间48min;在此条件下总黄酮的提取率为3.116%。响应面优化得到的提取工艺稳定合理,准确可靠,是提取八宝景天叶中总黄酮的可行方法。Abstract: Response surface analysis methodology was used to optimize the extraction technology of total flavonoids from Sedum spectabile Boreau leaves.The total flavonoids were extracted using ultrasonic assisted ethanol extraction.On the basis of single factor, Box-Behnken response surface methodology was employed for selecting the optimum extraction process by the indexes of the contents of total flavonoids from Sedum spectabile Boreau leaves.Total flavonoids of optimum extraction conditions were as follows, 72% of the ethanol concentration, liquid-solid ratio of 21:1, ultrasonic temperature of 64℃, ultrasonic time 48min.Under these conditions, the total flavonoids extraction was 3.116%.The extraction process obtained by response surface methodology was stable and reasonable, accurate and reliable.It was a feasible method to extract the total flavonoids from Sedum spectabile Boreau leaves.
